Progynon-Depot contains estradiol valerate, a synthetic analogue of the natural female sex hormone estradiol. It is used in hormone replacement therapy (HRT) to compensate for estrogen deficiency in women.
Indications for Use:
- Menopausal Syndrome:
- Relief of symptoms associated with estrogen deficiency in peri- and postmenopause, such as hot flashes, sweating, sleep disturbances, irritability, and more.
- Prevention of Osteoporosis:
- For postmenopausal women at increased risk of fractures, particularly if other medications are contraindicated or poorly tolerated.
- Hypoestrogenic Conditions:
- Due to hypogonadism, castration, or primary ovarian insufficiency.
Contraindications:
- Known hypersensitivity to estradiol or any components of the drug.
- Pregnancy and Breastfeeding.
- Estrogen-Dependent Tumors: Confirmed or suspected (e.g., breast or endometrial cancer).
- Unexplained Vaginal Bleeding.
- Acute or Chronic Liver Diseases.
- Thromboembolic Diseases: Active or with a history of conditions such as deep vein thrombosis or pulmonary embolism.
- Severe Hypertension.
Side Effects:
- Endocrine System:
- Changes in body weight, fluid retention, altered libido.
- Nervous System:
- Headache, dizziness, depressive states.
- Digestive System:
- Nausea, vomiting, abdominal pain.
- Skin:
- Skin rash, pruritus (itching), chloasma (pigmentation changes).
- Cardiovascular System:
- Elevated blood pressure, thromboembolic complications.
- Other:
- Soreness or tightness at the injection site.
Special Instructions:
- Pre-Therapy Examination:
- A comprehensive medical evaluation is required before initiating therapy, including a gynecological exam and mammography.
- Regular Monitoring:
- Periodic medical examinations are recommended during treatment to assess the patient’s condition and evaluate therapy effectiveness.
- Thromboembolism Risk:
- In women with an increased risk of thromboembolic complications, carefully weigh the benefit-to-risk ratio before prescribing.
- Laboratory Interference:
- Estrogens may influence the results of certain laboratory tests. Inform medical personnel of the medication use before testing.
- Consultation Required:
- Before starting Progynon-Depot, consult a healthcare provider to determine the appropriateness of treatment and the optimal dosage. Self-medication is strongly discouraged and may lead to adverse outcomes.
- Solution for intramuscular injection.
- Ampules of 1 ml containing 10 mg of the active substance dissolved in an oil base (usually benzyl benzoate and castor oil for prolonged action).
- The depot form provides a long-term effect of the drug after a single injection.
